She asked to call us to call her by her nickname: くぅちゃん。 Somehow it's different than: くーちゃん。With my extremely limited Japanese skills, I can't tell the difference.

 

I'd assume it's more of a trailing off sound rather than an elongated vowel which would normally be represented by a full size vowel (ie. くう) in hiragana or the chōonpu (ie. クー) more typically used in katakana. Either that or it's some sort of emphasis on the pronunciation which I think some dialects make use of.
